ANTIFUNGAL LIPOSOMAL PREPARATIONS AND THEIR INFLUENCE ON GENE MET3 EXPRESSION IN THE BIOFILM OF of Candіda albіcans

Lots of fungal infections associate with the formation of biofilms which can resist to antimicrobial agents. Due to the extremely high resistance of biofilms to the antimicrobial preparations the principles of antimicrobial therapy should be significantly changed. New approaches to the treatment of infections occurring with the formation of biofilms could include the development of new formulations of antimicrobial drugs based on nanotechnology. The purpose of the given work was development of the new liposomal antifungal preparations and research of change the gene expression in biofilms of Candіda albіcans. In this study we investigated the expression of the genes connected with formation of fungis biofilms and in particular gene MET3 during of the action of antifungoid preparations and their liposomal forms on early stages of biofilms formation. Serial dilutions of terbinafine were prepared in culture medium and one hundred microlitres of each dilution were added to wells of a microtitre plate with Candіda albіcans biofilms and incubated within 24 hours. After that biofilms washed from preparations and determined the presence RNA which is transcribed from gene МЕТ3 that specifies on expression this gene. The action of terbinafine and and its liposomal form on the biofilm leads to the inhibition of gene expression to what absence RNA testified synthesized with the given gene. While at the control (in absence of antifungal preparations) was present RNA. The using of liposomal terbinafine led to reduction of minimum inhibitory concentrations at 8-12 time in comparison with the intact terbinafine and more effective of the inhibition of biofilms activity. The efficiency of the liposomal terbinafine depended on the structure and the charge of liposomes. The most effective were the negatively charged liposomes. Obtained data have shown, that the using of the negatively charged liposomal antifungal preparations obtained from the polar lipids are highly effective concerning of fungal infections caused by Candіda albіcans and testifies to the opportunity of their using in treatment of skin forms of a candidiasis.
